Activity-regulated cytoskeletal protein (Arc) is an.effector immediate early gene involved in consolidation of long-term memory, Since activation of glucocorticoid receptors is prerequisite for memory consolidation, we therefore aimed to study the effect of acute restraint stress on Arc gene expression in adrenalectomized rats. Restraint stress produced a significant and immediate increase in Arc gene expression in the medial prefr ontal cortex, but not in the parietal cortex or in the pyramidal cell layer of the hippocampus. This was surprising because novelty has been shown to produce an increase in the hippocampus,. We speculated if the regulation of Arc gene expression was dependent on the nature of the stressor and modulated by circulating glucocor ticoids. The basal level of Arc mRNA in adrenalectomized animals was high in the prefrontal cortex and no change was achieved after stress in these animals.
