1. Viability assessments of nonstressed cells using the fluorescent stains oxonol, berberine, and MgANS (magnesium salt of 8-anilino-1-naphthalene-sulfonic acid) were not significantly different from slide count, the current ASBC-recommended method. 2. Analysis of the viability of stressed cell populations using each fluorescent stain indicated a lesser degree of correlation to slide culture than when nonstressed cells were examined. 3. A comparison of mean variation indicated that the fluorescent stain oxonol produced results that were most similar to the 'expected' values for nonstressed and stressed cell populations. 4. When compared with the 'observed' viability (as determined by slide culture), berberine was the most accurate stain for analyzing nonstressed cell populations, while MgANS was the most efficient for stressed cultures. 5. In terms of interlaboratory reproducibility and intralaboratory repeatability, oxonol was statistically the best method overall. However, when compared with slide culture, MgANS produced the most reproducible and repeatable results for the analysis of stressed cells.
