Objective: The C57BL/6J mouse as a model of Diet-Induced obesity (DIO) exhibits the biological features of obesity and mild hyperglycemia. To evaluate this model, we measured gene expression levels associated with glucose and lipid metabolism. Design: 6-week-old male mice were fed ad libitum a high-fat diet (D12492; Research Diets, Inc.), or a normal diet (D12450B; Research Diets, Inc.) for 12 weeks. Blood biochemistry and gene expression level in epididymal white adipose tissue and liver were analyzed at 18 weeks of age. Result and discussion: Lep and Ucp2 expression levels were significantly increased in white adipose tissue of high-fat diet group in comparison with normal diet group. In contrast, Adipoq, Hsdllbl and Pparg expression levels were significantly decreased in white adipose tissue of high-fat diet group. We found that the increased expression of Lep was consistent with increased serum leptin value. And the decreased expression of Adipoq was consistent with decreased serum adiponectin value. In addition, the decreased expression of Hsdllbl and Pparg may be related to development of insulin resistance. In the liver, expression of genes associated with cholesterol metabolism(Hmgcr) and lipogenic enzyme(Fasn) was significantly decreased. The decreased expression of Hmgcr may be related to increased serum TCHO value. And expression level of Fasn was significantly decreased, but serum NEFA and TG values were not different between high-fat diet group and normal diet group. Thus, serum NEFA and TG values were hardly altered, but expression level of various genes required for fatty acid and triglyceride synthesis may be altered in C57BL/6J DIO model mice.
