Some Factors Affecting In Vitro Development of Date Palm (Samany) Plantles and Molecular Characterization

Effects of activated charcoal (AC) and liquid half MSm medium with the substrate perlite on the in vitro development of date palm cultivar Samany were studied. The results indicated that 88.33% out of 85% of the developed embryos could be regenerated to obvious shootlets on medium supplemented with AC after 60 days. While 11.11% out of 45% of embryo like structures could be emerged and developed to structures-like regenerates after 60 days on AC free medium. Addition of AC to the medium led to increase in root numbers, root length and shoot length; 0.63 root, 1.53 cm and 2.24 cm per month of incubation, respectively, higher than those on AC free medium which gave 0.45 root, 0.81 cm and 1.43 cm, respectively. It was found that, the best medium for developing the date palm plantlets in vitro was liquid half MSm medium with the substrate perlite giving the highest scores for number of roots, root length and shoot length; 1.85, 3.06 cm and 5.7 cm per month of incubation, respectively. SDS-PAGE protein profiles showed that the tested samples reflected the similarities between the parental offshoot and their regenerates showing protein homogeneity. This revealed the identity of the genetic background of the regenerates and their offshoot parent.
