复元醒脑汤对糖尿病脑梗死大鼠脑组织血流量及含水量影响的实验研究

         

摘要

Objective To study the effect of Fuyuan Xingnao Decoctio on brain water content and cerebral blood flow of diabetes complicating cerebral infarction. Methods SPF SD male rats were randomly divided into 2 groups: normal group (72=24) and diabetic model group (n=96). Diabetes mellitus after injecting untreated, high fat diet for 2 weeks later, the diabetic rats were randomly divided into diabetic cerebral infarction group, sham operation group, model group and treatment group. Mold making after gastric gavage, the treatment group of Fuyuan Xingnao Decoction daily by gavage, normal control group, diabetic group, sham operation group, model group for volumes of saline gavage for 3-7 d. 2 times of gastric gavage each day. Record the degree of recovery of neurological signs score everyday; the last after intra-gastric administration of glucose, were used to detect the rats, blood flow to the brain. Results Compared with sham operation group, diabetic group, normal group, the cerebral blood flow of the model group was decreased significantly (P<0.01), while the neurological behavior symptoms scores was decreased (P<0.01), the brain tissue water increased (P<0.01), blood - blood brain barrier permeability was increased (P<0.01). Compared with the model group, the treatment group showed improved the neurological signs defects, decease in cerebral water content and blood - brain barrier permeability, increase in cerebral blood flow. Conclusions (1) The experiment using autologous clot method on the basis of SD rat diabetes established the model of diabetic cerebral infarction. The model is associated with human primary brain infarction mechanism relatively. (2) Diabetes mellitus cerebral infarction rat were found that the cerebral tissue water,blood - brain barrier permeability and cerebral blood flow were decreased. (3) Fuyuan Xingnao Decoction has the effect on improving diabetic cerebral infarction rat postoperative neurological signs defect, and reduces the water content in brain tissue and blood brain barrier permeability and increases cerebral blood flow.%目的 观察复元醒脑汤对糖尿病脑梗死大鼠脑组织血流量及含水量的影响.方法 选用SPF级SD雄性大鼠120只,随机分为2组:正常组(24只)、糖尿病模型组(96只).糖尿病造模成功后不进行治疗,高脂饲料喂养2w后,将糖尿病大鼠随机分为糖尿病组、脑梗死假手术组、糖尿病脑梗死模型组、糖尿病脑梗死治疗组4组.造模完成后分别灌胃,治疗组每日以复元醒脑汤灌胃,正常组、糖尿病组、假手术组、模型组予等容量的生理盐水灌胃,12次/d,连续3~7 d.每天对其恢复程度进行神经体征评分;末次灌胃后,分别检测脑组织血流量指标.处死动物后检测各组大鼠的脑组织含水量、脑血管通透性.结果 模型组与假手术组、糖尿病组、正常组比较,神经缺损行为体征评分升高(P<0.01),脑组织含水量增加(P<0.01),血-脑屏障通透性升高(P<0.01);治疗组与模型组相比,神经体征缺损明显改善(P<0.01)、脑含水量、血-脑屏障通透性降低(P<0.01),梗死侧脑血流量增加(P<0.01).结论 (1)本实验在SD大鼠糖尿病的基础上采用自体血栓法建立糖尿病脑梗死模型,该模型是与人类原发脑梗死的机理较为接近的动物模型;(2)糖尿病脑梗死大鼠出现脑组织含水量增加、血-脑屏障通透性升高、脑血流量下降等现象;(3)复元醒脑汤对糖尿病脑梗死大鼠术后神经体征缺损具有明显改善作用,并降低了其脑组织含水量和血-脑屏障通透性,提高了脑血流量.
